The hotel will be located in front of the Potawatomi Bingo Casino in Milwaukee.
It will feature 381 rooms, a spa, a restaurant, a bar/lounge and meeting space.
Construction will generate nearly 1,000 jobs. The tribe will hire 230 people for the hotel, which is expected to open in summer 2014.
